Technology of the week: DNA-Encoded Libraries
Chemical space is theoretically >1x10^26
- Did you know that you can test 1x10^16 candidates against one target in a single tube? How? DNA-encoded Libraries.
A DNA-encoded library is a collection of small molecules covalently linked to DNA that has unique information about the identity and the structure of each library member.
A DNA-encoded chemical library (DEL or DECL) is broadly adopted by major pharmaceutical companies and used in numerous drug discovery programs like:
- X-Chem, Inc. (250 billion compounds per screen)
- Nurix Therapeutics
- DyNAbind GmbH
- WuXi AppTec China (90 billion compounds per screen)
- Enamine Ltd.
- Amgen
- Roche
- Takeda
- Sanofi
And many more…
Here is a wonderful introductory publication to get acquainted with the technology. This publication is by Robert Goodnow (Takeda) Christoph Dumelin (Novartis) and Anthony Keefe (X-Chem, Inc.)
