Technologies of the week: CVLs and DNA Data Storage

With the ability to print DNA base-by-base, we can use human scaffolds/domains as inspiration or a starting point when developing a drug discovery/screening campaign or when trying to understand what happens in the mutated states of a specific protein.

With Twist Bioscience's Combinatorial Variant Libraries (CVL) you can create/optimize synthetic antibodies, synthetic virus, B-cells, TCRs, etc. with the precise ability to design custom libraries of particular domains in each of these biologics.

“While working with Twist in 2020-2021 to develop synthetic antibodies against SARS-CoV-2 S1 protein and ACE-2 receptor, we used CVLs to remove glycosylation sites and/or deamidation sites that would interfere with expression while, simultaneously, humanizing our synthetic antibody candidates through hyper-diversification of CDR domains which helped mimic the human repertoire (in attempt) to evade immunogenic reactions in clinical settings.” - Thomas Perkins, Jr.

Another insane application CVLs spans into is DNA Data Storage. The ability to discreetly print a corresponding nucleotide for every binary pair received in a dataset.
